Subclinical mastitis is a kind of mastitis which is difficult to be detected by farmer, because the symptom can’t be visually observed. One detection method of subclinical mastitis which is known to be simple and quick is CMT method. But, this method is poorly sensitive in deciding the severity of subclinical mastitis, so it needs a further molecular research. IL-8 is a one of essential molecular marker in mastitis pathogen elimination process because IL-8 can activate and migrate neutrophil into the infection area to destroy the pathogen. This is related with the function of IL-8 in cell as chemoatractan and main activator of neutrophil. This research aimed to show the profile of IL-8 protein expression in subclinical mastitis cow and evaluating the relationship between IL-8 concentrations with mastitis score according to CMT. Blood and milk samples in this research was obtained from 30 Friesien Holstein cows (PFH) (120 nipples) that belonged to KUTT Suka Makmur Kecamatan Grati Kabupaten Pasuruan. Those samples then were tested for IL-8 protein profiles through SDS-PAGE and the confirmation of IL-8 through imunohistochemistry method in blood smear. On other hand to decide IL-8 concentration, a counting ofcomplex bond absorbance of IL-8 and anti IL-8 in ELISA at 450 nm wavelenght were carried out. The result of research showed that from 30 dairy cows samples, there were 13 (52 nipples) which could be classified as normal according to CMT (zero score) and the rests were suffering from subclinical mastitis with score CMT 1-4. Then, the observation of IL-8 protein profile showed that IL-8 was a protein with 8,5 kDa molecular weight appeared in all of CMT scores. The IL-8 concentration positively correlated with mastitis score with determination coefficient of 99,5%. So, it can be concluded that higher mastitis levels of a dairy cow has higher IL-8 concentrations (JIIPB 2008 Vol 18 No 1 : 36-50).Keywords: Mastitis, IL-8, CMT score and Dairy Cattle
