μCurrently, the use of lingzhi (reishi, Ganoderma lucidum) for curing some diseases had increased. Preleminary phytochemical screening of lingzhi showed that its contents were alkaloid, flavonoid, saponin, quinone, steroid/terpenoid, and coumarin. Maceration of dried slices of lingzhi with deionized water, ethanol, and acetone obtained water extract (87.3 mg, 4.37%), ethanol extract (296 mg, 1.48%), acetone extract (18.7 mg, 0.94%) and ethyl acetate extract (26.2 mg, 1.31%), respectively, after the filtrates were concentrated using rotary evaporator followed by drying in oven desiccator in vacuo. Cytotoxic activity test against leukemia L1210 cells of those extracts showed that the cytotoxic activity of ethanolic extract (IC50=18.1μg/ml) was the strongest. Further separation of ethanolic extract by solvent partition into water-chloroform (1:1) followed by cytotoxic ativity test against leukemia L1210 gave the IC50 value of 28.2 ?g/ml for water phase and 17.6 ?g/ml for chloroform phase. The separation of chloroform phase using column chromatography method with gradient solvent system resulted five fractions, Fr-1 to Fr-5. The cytotoxic activity test of the five fractions against leukemia L1210 cells showed that IC50 values of those fractions were between 10.5 up to 19.9 ?g/ml, meaning that all fractions demonstrated potential anticancer agents which had IC50 value less than 20 ?g/ml. Among the fractions, Fr-1 was the most potential with IC50 was 10.5 μg/ml and had highest yield of 25.7 %.
